2. Which layout best fits a FormPanel when the fields need a complex arrangement e.g first row contains a single textbox whereas the second row contains 2 text boxes

You could specify a larger labelWidth config, which would move the field further right.
Re 1: AFAIK, label to field spacing is not configurable. I'd think that you could override some css classes to alter that spacing. Explore rendered form html to find out exact css class names that could be influencing that spacing. Re 2: I'd use column layout for that item where you want two text fields in one row.
